Polymer microfluidic chip with embedded CNTs for DNA sample preparation

We present a polymer-based microfluidic chip which has deposited carbon nanotubes (CNTs) onto the microchannel. The chip is capable of performing DNA preparation process like restriction enzyme digestion and ligation of DNA. The advantage of having a large surface area to volume ratio from deposited CNTs in micro channel introduces high binding capacity of DNA. CNTs on polymer channel were coated with a copolymer (PEG-PEI) and were used to immobilize DNA via streptavidin-biotin interactions. Integration of two enzymatic reactions into a single microfluidic polymer chip was performed in this work. The chip was associated with automated control system. Biotinylated DNA was first immobilized on the surfaces of CNTs. The amount of DNA bound onto the chip was quantified by a Picogreen test after collecting the supernatant. Immobilization of DNA onto CNTs chip was achieved up to 190ng DNA. DNA which was immobilized on chip was further digested with Hind III. Restriction digestion process was performed at 37°C for 2 hours. Supernatants from chips were run on gel electrophoresis to qualify digested DNA. Gel electrophoresis results show that restriction digestion of DNA was successful as the correct DNA band size was observed on the gel.
