Polymersomes as model of transcytosis across Blood-Brain Barrier (BBB)

In this work, two formulations kinds of diblock copolymers ((poly(2- methacryloyloxy)ethyl phosphorylcholine)-co-poly(2-(diisopropylamino)ethyl methacrylate(PMPC25-­‐PDPA70) and poly(ethylene glycol)-co-poly(2- (diisopropylamino)ethyl methacrylate)(PEO113-­‐PDPA70))will be employed,blending them in series ratios in order to obtain the best morphology and size across the blood brain barrier. The primary study focus on the uptake kinetics and fast screening of Polymersomes; based on the screening result, best formulation will be used to tailor make BBB targeting polymeric nanoparticles. This will be done using transcytotic-­‐receptor modified polymersomes to increase uptake kinetics enhancing also differential uptake by BBB endothelial cells, achieve the cellular uptake by transcytosis.
