Invention of Polysaccharide-base Nanoparticles for Enhancing Drug Permeability across the Blood Brain Barrier

The primary objective of this study is to evaluate novel cationic and degradable beta-cyclodextrin nanoparticles as drug delivery carriers for insulin across the bovine brain microvessel endothelial cell (BBMVEC) monolayer, an in vitro blood brain barrier (BBB) model. We use dynamic light scattering, Zeta potentiometer and FTIR to characterize the particle sizes, zeta potentials and degradation of the nanoparticles. We study the release kinetics of insulin from the nanoparticles. We investigate the permeability of the nanoparticles without/with Doxo and insulin across the BBMVEC monolayer. We also assess the in vivo biodistribution of the nanoparticles. Our preliminary results demonstrate that the nanoparticles can sustain release of insulin, and are not only twice as permeable as 4 kD dextran control, but also enhance insulin permeability across the BBMVEC monolayer.
