Nanobiopore: A Novel Nano-Porous Electrode System to Enhance Biosensor Sensitivity

The primary objective of this research is to create electrode systems for use as electrochemical transducers with enhanced sensitivity in order to enable detection of low concentration diagnostic targets not accessible to date. NanoBioPore sensors consist of micro- or nano-porous metal-insulator-metal layer systems and exhibit an effective electrode spacing of only 100 nm. Patterning of pores was achieved by utilizing self-assembled particles as shadow masks during the deposition of the top electrode. Amplification factors > 50 have already been demonstrated, which is a significant improvement when compared to redox cycling factors reported from measurements using conventional interdigitated electrode (IDE) systems. A variety of biomolecules conjugated with alkaline phosphatase were immobilized in the pores and shown to be functional as evident from detection of p-aminophenol generated by the enzyme. In conclusion, high sensitivity combined with low fabrication costs due to the utilization of nanosphere lithography should open a broad range of applications for these sensors, particularly in point-of-care diagnostics.
