Microfluidic Valve-Controllable Magnetic Bead Receptacle: A Novel Platform for DNA Sequencing

In this paper, a novel and effective microfluidic platform for DNA Sequencing, particularly for sequencing-by-synthesis methods such as Pyrosequencing, based on valve-controllable PDMS channel system contains micrometer magnetic beads . Additionally, the proposed picoliter micofluidic system is essentially useful for a number of other bio-species detection and sorting templates . The encapsulation of bead and reaction in picolitre-sized vessels gives an excellent isolation and control on the detection based system and prevent cross-talk among neighbor reactors very well. In addition, electromagnetic control on the bead’s location and movement eliminates flow and washing issue in the system. In conclusion, it leads to a much higher sensitivity platform in real time than the current state-of-the-art technology. The design and fabrication of the platform followed by experimental results of the initial system are presented. The method is subsequently used to explore the impact of the design’s shape and dimensions of microchannels, fluidics parameters, leaky valve trapping mode and the role of biotinylated DNA attached on the beads and finally the resultant potential tradeoffs are discussed which show the advantages of this method in practical applications.
