High throughput leukemia cell(k-562) sorting system based on negative dielectrophoretic force

For high throughput and high separation efficiency, the proposed sorting platform comprises meso-size vertical channel and silicon wafer based micro cantilever-type electrodes for generating n-DEP force and playing a role of micro-channel. To reduce engineering cost, we adopt the flow regulator to generate gravity based hydrodynamic force instead of microsyringe pump. In the proposed platform, therefore, live cells and dead cells are exposed to gravitational force, hydrodynamic force. Then, the n-DEP force generated from electric field selectively acts on the cells according to signal condition. As a consequence, the direction of cells is determined by the vector summation of among gravitation, hydrodynamic force and n-DEP force.By investigating magnitude of each force according to operation conditions such as voltage level and frequency, flow rate, and conductivity of medium, we tried to find optimized experimental conditions to achieve the best separation efficiency. Conclusively, the target cells are effectively separated under the condition of voltage 7Vpp, frequency 100kHz and flow rate of 10uL/min and 20uL/min in the channel 1& 2 respectively. Under these conditions, experimental efficiency was observed 87% and 82% in the channel 1 and 2 respectively. In conclusion, we achieved high throughput of 2400cell/sec with high separation efficiency of 86%.
