| Abstract: | Gold-gold sulfide nanoparticle (GGS-NP) with controllable near infrared (nIR) absorption was synthesized from a simple reaction of chloroauric acid and sodium thiosulfate. Through PEGylated recombinant Protein-G (ProG), ProG-conjugated GGS-NPs for antibody capture by Fc domain provided a practical stratagem for early-stage cancer diagnosis and targeted therapy. In vitro studies, the as-fabricated ProG-conjugated GGS-NPs with bound anti erbB-2 (HER-2) IgGs successfully targeted at HER-2 overexpressing breast cancer cell, SK-BR-3, and killed them at an optimized low laser power. The concept of the targeted photothermal cancer therapy using ProG as a cofactor for immobilization of antibody is well proved and very promising as provides a repeatable platform method for antibody conjugation to gold nanoparticles. |
