Phosphate recognition using multilayer charged thin films containing zirconium (Zr) ions for biochip applications

Abstract:The phosphate residue plays an important role as not only the structural component of DNA and lipid but also the activation signal of protein in the cellular signaling pathways. Therefore, the selective recognition for the phosphate group is very important in the field of biochip and biosensor technologies. We have focused on these background and possibility of crystalization between phosphate and zirconium (Zr). To fabricate phosphate specific sensing membrane, we constructed a multilayer charged thin (MLCT) film containing zirconium (Zr) and evaluated its ability to recognize the phosphate group. Oligonucleotide fragment terminated with phosphate and fluorescent dye (Cy5) were used as a probe for evaluation of phosphate recognition ability. In this work, our results indicate that the multilayer charged thin (MLCT) film containing zirconium (Zr) can recognize phosphate group selectively. In addition, through the estimation of fluorescence intensity due to immobilized oligonucleotide probes, we had tried to optimize the fabrication conditions of sensing membrane with respect to the polymer content, dipping time, and repeatation number of layer. Taken together, these results suggest that the multilayer charged thin (MLCT) film containing zirconium (Zr) can be used for the specific recognition of biomolecules containing phosphate group and also for the application toward the phosphate-specific biochip and biosensor.
