One-step Target Protein detection from Whole Blood in a Lab-on-a-Disc

Abstract:We report a fully integrated, one-step Lab-on-a-Disc for detecting target protein from whole blood utilizing centrifugal microfluidics, so called “Immunoassay Lab-on-a-Disc”. The design principle is the microbead-based sandwich ELISA on the Disc. The microbeads are conjugated by target specific antibody or antigen covalently on their surface, so that they can capture the target proteins efficiently by mixing. As model studies, the detection of Hepatitis virus B surface antibody (Anti-HBs) and antigen (HBsAg) in whole blood were conducted. Total process starts automatically after loading 120 l/test of whole blood and is finished within 38 min. Except for loading whole blood sample, all kinds of reagents are preloaded. Compared to the traditional ELISA method, the Lab-on-a-Disc method can reduce the total operating time dramatically (traditional ELISA 150 min, Lab-on-a-Disc 38 min) with the same functional sensitivity (10 mIU/ml for Anti-HBs, 0.5 ng/mL for HBsAg). The Immunoassay Lab-on-a-Disc itself can provide a general platform to detect various kinds of proteins.
