Abstract:Poly(ethylene glycol)-Poly(?- caprolactone)-Poly(?-amino ester) (PAE-PCL-PEG-PCL-PAE) - a novel pH/temperature sensitive and biodegradable hydrogel copolymer used for drug and protein delivery was synthesized. Bovine insulin (BI) and human growth hormone (HGH) loaded well to the copolymer to form a complex mixture by ionic link. The releasing of BI and HGH was investigated in both in vitro. The results show that BI which was loaded in the polymer was released within 13 days, and the releasing time of HGH was 10 days. The complex mixture of human insulin (HI) and copolymer was injected subcutaneously through a needle to rat, and diabetic rat by in vivo. In conclusion, PAE-PCL-PEG-PCL-PAE seems to be useful as a biodegradable and subcutaneously injectable protein and drug carrier.
