Synthesis and differential uptake of drug-decorated fluorescent nanoparticles by live cancer cells

Abstract:Design and applications of “smart” multivalent fluorescent probes based on semiconductor nanocrystals and anticancer drug molecules will be presented. Various polymer coating strategies have been employed to insure stability and define composition of nanoparticles. Next, a series of drug-functionalized polymer coated nanoparticles with various drug loadings have been synthesized and their uptake by corresponding drug-sensitive and drug-insensitive cancer cells have been studied. We have observed a strong correlation between nanoparticle uptake and the number of drug molecules attached to the nanoparticle. This is the first example of drug-mediated uptake of nanoparticles by live cells. Synthesis and characterization studies for a number of multivalent nanoparticles where drug was used in a combination with intracellular transporter moieties are ongoing. Physical and chemical properties, stability and composition of nanoparticles have been studied by TEM, DLS, and UV-visible and fluorescence spectroscopic methods. Potential applications of this technology include specific labeling of live cancer cells, studies on a mechanism of drug action, early cancer diagnostics and drug delivery.
