Cytotoxicity and genotoxicity of silver nanomaterials

Abstract:

Summary We have investigated the cytotoxic effect of dialyzed silver nanoparticles (Ag NPs), PEGylated Ag NPs and silver nanorods on human cultured cells. The cytotoxicity of 24 h treatment with Ag NPs at 0.25, 0.5, 1.0, 5 and 10 ug/cm2 were as follows: 97.5%, 91.7 %, 81.8 %, 27.8% and 24.7% relative survival, respectively, for umbilical vein endothelial cells HUVEC; 99%, 95.6%, 92%, 0.11 % and 0% relative survival, respectively, for T leukemia cell line JURKAT; and 97.3%, 96%, 87.5%, 65% and 51.8% relative survival, respectively, for human PBMC. The Ag NPs showed higher toxic effect at longer exposure time, 120 h: 63.7%, 57.63 %, 34.23 %, 1.03 % and 0 % relative survival were found in HUVEC treated with Ag NPs at 0.250, 0.5, 1.0, 5 and 10 ug/cm2,respectively. Such treatment resulted in no survival for JURKAT and B lymphoblastoid cell line RAJI. PEGylated Ag NPs appeared to be less toxic to JURKAT. Among the 3 types of silver nanomaterials, silver nanorods were found to have higher toxic effect on the human cell lines tested. Using the cytokinesis-block micronucleus assay we also demonstrate that Ag NPs can cause DNA damage in human peripheral blood cells. Topic area: Toxicology
