Selection of SKBR-3 breast cancer cell-binding and internalizing peptides by landscape phage display

Abstract:
We used landscape phage library to identify SKBR-3 cell binding peptides. New SKBR-3 binding peptides, including DGSIPWST, VSSTQDFP, DNGSGISW, DTKTAPAW, AFSEAAQT, GSTDSDLK, ADAPSNGW, ATTAPSYP, AAATKSDL and DNDNYAFP, were selected. Among them, DGSIPWST and VSSTQDFP were found to be the best cell-binding peptides that are also the cell-internalizing peptides. The affinity-selected peptides were analyzed by using REceptor LIgand Contacts (RELIC) program, which is a bioinformatics tool for combinatorial peptide analysis and identification of protein ligand interaction sites. The cell-specific binding of affinity selected peptide was further confirmed by ELISA and immunofluorescence imaging.
