Influence of Ultrasonic Waves on Escherichia Coli DH5a Genom

Abstract:

The effect of 1 MHz ultrasound on E. coli DH5alpha during water disinfection process was investigated. During ultrasonic irradiation of water, OH- and H+ products are formed (Mason et. al., 1994; Suslick, 1988). Bactericidal properties of free radicals act directly to DNA cells. Ultrasound waves produces cell lysis, membrane impairment and DNA degradation on bacteria. Ultrasonic vibration causes DNA fragmentation and free radicals attack water bonds of DNA fragments (Hughes and Nyborg, 1962). 1 MHz ultrasonic treatment was applied to water samples during 1, 2, 5 and 10 min. Results are shown in figures 1 and 2. Figure 1 represents the results for the electrophoresis gel of the treated samples. The electrophoresis gel doesn’t show evidence of DNA degradation as a result of the ultrasonic irradiation to water samples containing Escherichia coli DH5alpha. However, figure 2 representing the gel after PCR with OPR primers procedure shows that ultrasound irradiation during 10 min affects the DNA structure.
