Subcellular fractionation in a fluidic microsystem

Abstract: The availability of an easy-to-use, reproducible method for subcellular fractionation would significantly promote qualitative and quantitative analysis of subcellular protein composition. A microfluidic system with integrated electrodes was designed and fabricated allowing for dielectrophoretic manipulation of biological nanoparticles. Fluorescently labelled mitochondria were successfully retrieved from a complex cell homogenate employing a dielectrophoretic band-bass electrode array. Dielectric properties of mitochondria were also investigated and biochemical analysis of fractions obtained was performed.
