Nanoscale imaging and mechanical testing of cells and tissues

Abstract:
Characterization of the structure and mechanical properties of cells and tissues at the nanoscale imposes a tremendous challenge to many existing imaging and mechanical testing techniques and instruments. AFM has been used to image dry and wet cells and tissues. Experimental and instrumentation difficulties in imaging are discussed. Nanomechanical testing techniques such as nanoindentation, axial tensile testing, and in-situ AFM tensile testing are presented. Viscoelastic property measurement techniques have been used to assess the storage and loss moduli and creep resistance. The technique for charactering fatigue properties is described. Instrumentation limitations and calibration techniques are discussed.
