The micro and nano scale characterization of polymer coated drug delivery stents

Abstract:
Several PLA/Everolimus stent samples have been analyzed under ambient and liquid conditions with a D3000 AFM (Veeco, CA). A rapid loss of the domain material from the stent surface has been found when exposed to buffer. This is a rapid process leaves pits in the surface. Over time (monitored up to 60 minutes) in buffer, the pits in the surface broaden and increase in depth, with some evidence of all domain material from the surface being lost by this time. On keeping the stent in buffer for several days, the polymer surface was observed to soften and for material to be apparently lost from the surface; clear depressions could be observed at 15 days exposure onwards. These data are beginning to indicate the possible release mechanism of the stents, showing a rapid initial erosion of the domain material (which may contain the drug) over the period of an hour or so and a much slower erosion of the matrix material over many days and months.
