Compact Disc (CD)-Based Automated Animal Assay

Abstract:
Microfabricated platforms that allow automated cultivation and behavioral observation of Caenorhabditis elegans would be a valuable tool in physiological and genetic studies. Here, we report an automated microfluidic compact disc (CD) system conducive to the cultivation and monitoring of C. elegans. This system allows automated feeding and waste removal, and live-animal microscopy for up to two weeks. In addition, the desired gravity levels can be obtained by manipulating the rotational speed and the geometry of discs. One purpose is to use 100xG hypergravity conditions as a novel stress paradigm to investigate the genetic basis of mechanical stress, the effects of mechanical tension on development, metabolic homeostasis, reproduction and ageing, and the interaction between mechanical stress and other forms environmental and physiological stresses. Another purpose is to investigate pharmacological agents that may modulate the stress responses.
