Integrated Techniques for Transmembrane Protein Sensing

Abstract:
We present a method for the microfabrication of apertures in a silicon substrate using well-known cleanroom technologies and a novel approach using phase sensitive detection to measure single channels of transmembrane proteins. These proteins are of considerable interest because they are fundamental to sending signals throughout the human body. Microfabrication of parallel apertures of such devices could be used for high throughput screening methodologies in both pharmaceutical and integrated biological sensor applications. Using engineered proteins and the presented integrated device, a sensor capable of detecting single molecular analytes can be envisioned. Our results demonstrate a working fabricated silicon device that is capable of such measurements. We also demonstrate a lock-in amplifier technique used to record single channels of transmembrane proteins inserted into bilayer membranes formed on the silicon device.
