A Simple and Direct Biomolecule Detection Based on a Microwave Resonator

We report a simple and direct biomolecule detection method, sensitivity enhancing technique, and its characterization realized by using a CPW-to-slotline ring resonator whose resonant frequency is 3.4 GHz. When a biomolecule is attached on the resonator, it directly detects a biomolecule through its resonant frequency change without an additional transduction procedure.
The microwave biosensor was fabricated and characterized through biotin-streptavidin reaction. The results show that its resonant frequency change was 65 MHz and 10 MHz due to biotin and streptavidin binding to the resonator, respectively.
To enhance sensitivity, the resonator was modified by increasing reaction area and utilizing a region where electromagnetic field intensity concentrated. Compared to resonant frequency change of the original resonator, that of the modified resonator showed sensitivity enhancement by 21% (79 MHz) and 80% (18 MHz)when biotin and streptavidin was bound.
According to the results of the proposed microwave biosensor, it is expected that the biosensor can simplify biomolecule detection process.
