Molecular Dynamics of Signal Peptides of NS2-protein of Hepatitis C Virus

Abstract:
NS2-protein of Hepatitis C Virus is a 23KDa protein. With N-terminal part of NS3-protein it forms serine protease that induces cleavage of NS2-NS3 protein. NS2-protein is transmembrane protein of 217 aa. It’s activity depends on it’s localization in membrane. NS2-protien contain four transmembrane domains, two of them are signal sequences. Inducing point mutations in these signal sequences may change it’s localization and, in consequence, it’s activity as a serine protease.
Molecular Dynamics simulations are used to determine what local mutations can radically change hydrophobic activity of NS2-protein.
